RBC Real Genomics™ 96-well Genomic DNA Kit is based on a reliable convenient silica format and is compatible with both centrifuge and vacuum protocols. It contains all the necessary plates, buffers and reagents for easy use straight from the kit. Fresh or frozen blood may be utilized. The volume per well is optimal at 200µl, yielding an average of 3-8µg which is sufficient DNA for many subsequent applications. The high quality silica membrane DNA filter plate ensures low well to well and batch to batch variability.

Blood  Samples

Fresh blood is best used within 1 week (Room Temp/40C). EDTA must be added to prevent coagulation. Blood sample characteristics may vary according to indvidual, age, health status, storage time etc. Mix samples gently by inverting before use. Avoid using clotted blood, check blood sample by visual inspection.  Clots will block the filter column. Do not allow to dry before applying to preparation plate. This protocol is optimized for 200µl samples.  If sample volume is less than 200µl, add PBS buffer to make up to 200µl. Max sample up to 400µl must be split and lysed in two separate  wells.  After lysis the lysate may be recombined and applied to single DNA Binding Well.

Centrifuge
Any Medium/High Speed Centrifuge (Beckmann/Eppendorf/Qiagen/Sigma) equipped with 2 x 96 Plate Rotor.

Vacuum System
RBC Multi-Well Vacuum Manifold or any standard 96/384 microplate vacuum manifolds may be utilised. The vacuum protocol is ideal for benchtop operation, enabling processing of just one plate at a time and a lower cost solution than centrifuging. RBC recommends a vacuum pressure starting from 10 inches Hg up to maximum 20 inches Hg to ensure all solution passes through each well. In practice all the wells should pass through solution within 3-5 minutes, the yields will show little variation even if some wells pass the solution through at varying speeds. Even if some wells process almost immediately following vacuum the DNA will still bind efficiently to the filter matrix. Maintain vacuum until the solution fully passes through each well of the plate.
