Real Genomics™ 96-well Plasmid DNA Kit is designed for rapid isolation of plasmid or cosmid DNA from 1- 2ml of bacterial cultures. It contains all the necessary plates, buffers and reagents for easy use straight from the kit. Centrifuge and combined Centrifuge and Vacuum Protocols are included. In the process, the modified alkaline lysis method  and RNase A treatment are used to obtain cleared cell lysate with minimal genomic DNA and RNA contaminants. The volume per well is optimal at 1- 2ml, yielding an average of 5-10µg which is sufficient DNA for many subsequent applications. The high quality silica membrane DNA filter plate ensures low well to well and batch to batch variability.

Quality Control
The quality of 96-well Plasmid Kit is tested on a lot-to-lot basis. The kits are tested by isolation of plasmid DNA from 1.5ml culture of E. coli DH5alpha, which contains the plasmid pUC19 (A600 > 2units/ml). More than 8mg of plasmid DNA was quantified with a spectrophotometer. 1µg of the purified plasmid was restriction enzyme digested with EcoRI and the digested DNA was checked by agarose gel analysis.
