The Fast Ion™ Plasmid Midi Kits use pre-packed anion exchange resin columns to purify plasmid DNA from bacterial cultures. In the process, the modified alkaline lysis method and RNase treatment are used to get cleared cell lysate with minimal genomic DNA and RNA contaminants. Following binding of the plasmid DNA to the column, the contaminants can be washed off with wash buffer. Finally, the purified plasmid DNA is eluted with a high salt buffer and precipitated with isopropanol for desalting. The entire procedure can be completed in 120 minutes resulting in high purity plasmid DNA.

Quality Control
The quality of Plasmid Midi/Maxi Kit is tested on a lot-to-lot basis. The kits are tested by isolation of plasmid DNA from 100ml cultures of E.Coli DH5alpha containing the plasmid pBluescript (A600>2 units/ml). More than 400µg of plasmid DNA was quantified with a spectrophotometer. 1µg of the purified plasmid was used for restriction enzyme digestion with EcoRI and DNA is checked by agarose gel analysis.
